Prednisolone is a short-acting synthetic glucocorticosteroid. It has anti-inflammatory, anti-allergic, anti-stress, gluconeogenetic action. It interacts with specific cytoplasmic receptors to form a complex, which penetrates into nucleus and induces formation of proteins, including enzymes regulating various processes in cells.
Prednisolone stimulates gluconeogenesis, increases glycogen accumulation, increases blood glucose level and stimulates insulin secretion, inhibits glucose capture by fat cells, which leads to lipolysis activation. It has a catabolic effect in the lymphoid, connective, fat and bone tissues, muscles and skin. It inhibits all phases of inflammation, suppresses functions of leukocytes and tissue macrophages, limits migration of leukocytes to the inflammation focus, prevents phagocytosis and interleukin-1 formation, helps stabilize lysosomal membranes, reducing the concentration of proteolytic enzymes in the inflammation focus. It reduces capillary permeability caused by histamine release, inhibits fibroblast activity and collagen formation. Antiallergic effect is caused by decrease in the number of basophils, direct inhibition of secretion and synthesis of histamine. Anti-shock effect is due to enhancement of catecholamines action and restoration of receptors sensitivity to catecholamines.
The drug is well absorbed from the injection site, evenly distributed in tissues, metabolized by liver, excreted with urine and bile.
Special instructions
Use with caution in pregnant women.
Do not mix with other drugs in the same syringe. When used simultaneously with thiazides and loop diuretics, potassium loss increases; when used simultaneously with nonsteroidal anti-inflammatory drugs, the risk of ulcers in the stomach and intestines and gastrointestinal bleeding increases; when used simultaneously with anticholinergics, intraocular pressure increases. Barbiturates, phenytoin and rifampicin reduce the glucocorticoid activity of prednisolone.

Side Effects
With long-term use, reversible atrophy of the adrenal cortex, immunosuppression, slow wound healing and callus formation, osteoporosis, arthropathy, muscle atrophy, growth retardation, diabetes mellitus, Cushing’s syndrome, pancreatitis may occur. In cats, there is excitement followed by depression, in dogs – periodic depression and aggression, skin atrophy, glaucoma, cataracts, polydipsia, polyphagia, polyuria, hypertension, ulcers in the stomach and intestines, reversible hepatopathy, thrombosis, sodium retention in the body and edema, hypokalemia, hypocalcemia, abortions in cows in the last third of pregnancy, temporary decrease in milk production productivity in cows.
Contraindications
Do not use in case of heart and kidney failure, hypercortisolism (Cushing’s syndrome), diabetes, osteoporosis, viral diseases and routine vaccinations.
