Prednisolone is a short-acting synthetic glucocorticosteroid. It has anti-inflammatory, anti-allergic, anti-stress, gluconeogenetic action. It interacts with specific cytoplasmic receptors to form a complex, which penetrates into nucleus and induces formation of proteins, including enzymes regulating various processes in cells.
Prednisolone stimulates gluconeogenesis, increases glycogen accumulation, increases blood glucose level and stimulates insulin secretion, inhibits glucose capture by fat cells, which leads to lipolysis activation. It has a catabolic effect in the lymphoid, connective, fat and bone tissues, muscles and skin. It inhibits all phases of inflammation, suppresses functions of leukocytes and tissue macrophages, limits migration of leukocytes to the inflammation focus, prevents phagocytosis and interleukin-1 formation, helps stabilize lysosomal membranes, reducing the concentration of proteolytic enzymes in the inflammation focus. It reduces capillary permeability caused by histamine release, inhibits fibroblast activity and collagen formation. Antiallergic effect is caused by decrease in the number of basophils, direct inhibition of secretion and synthesis of histamine. Anti-shock effect is due to enhancement of catecholamines action and restoration of receptors sensitivity to catecholamines.
The drug is well absorbed from the injection site, evenly distributed in tissues, metabolized by liver, excreted with urine and bile.

Composition

The preparation contains 0.025g of prednisolone, excipients and solvent up to 1.0 cm³.

How to take, the dosage

The drug is administered intramuscularly (preferably in the morning hours) in doses:

• to cattle, horses: 4 cm³/250 kg of animal weight (0.4 mg prednisolone/kg of animal weight);
• to sheep, goats, pigs, calves: 0.4 cm³/10kg animal weight (1mg prednisolone/kg animal weight);
• dogs, cats: 0.1-0.4 cm³/5kg animal weight (1mg prednisolone/kg animal weight).

If necessary the drug is repeated in 24 hours until recovery.
Abrupt termination of treatment with prolonged use may lead to exacerbation of the pathological process, so the dose should be reduced gradually.

Special Instructions

Use with caution in pregnant women.
Do not mix in the same syringe with other drugs. Concomitant use with thiazides and loop diuretics increases potassium loss; concomitant use with nonsteroidal anti-inflammatory drugs increases the risk of gastric and intestinal ulcers, gastrointestinal bleeding, concomitant use with cholinolytics increases intraocular pressure. Barbiturates, phenytoin and rifampicin reduce glucocorticoid activity of prednisolone.
Personal precautions
Personal hygiene and safety rules should be observed when handling the drug.

Contraindications

Do not use in cardiac and renal failure, hypercorticism (Cushing’s syndrome), diabetes, osteoporosis, viral diseases and scheduled vaccinations.

Side effects

With long-term use reversible atrophy of the adrenal cortex, immunosuppression, slow wound healing and bone callus formation, osteoporosis, arthropathy, muscle atrophy, growth retardation, diabetes, Cushing’s syndrome, pancreatitis may occur. In cats there is agitation followed by oppression, in dogs there is intermittent oppression and aggression, skin atrophy, glaucoma, cataracts, polydipsia, polyphagia, polyuria, hypertension, gastric and intestinal ulcers, reversible hepatopathy, thrombosis, sodium retention and edema, hypokalemia, hypocalcemia, abortion in cows in the last third of pregnancy, temporary reduction of milk productivity in cows.
