Interleukin-2 is produced by a subpopulation of T-lymphocytes (T-helper I) in response to antigenic stimulation. The synthesized IL-2 affects T-lymphocytes, increasing their proliferation and subsequent synthesis of IL-2. The biological effects of IL-2 are mediated by its binding to specific receptors present on various cellular targets. IL-2 directly affects the growth, differentiation and activation of T- and B-lymphocytes, monocytes, macrophages, oligodendroglial cells, epidermal Langerhans cells, increases the synthesis of all immunoglobulin isotypes by plasma cells. IL-2 reduces the level of spontaneous apoptosis of helper T-lymphocytes, increases the production of interferons α, β, γ. The development of cytolytic activity of natural killer and cytotoxic T-lymphocytes depends on its presence. IL-2 causes formation of lymphokine-activated killer cells and activates tumor-infiltrating cells. Expansion of the spectrum of the effector cells lysing action causes elimination of a variety of pathogens, infected and malignized cells, which provides immune protection directed against tumor cell growth, as well as viral, bacterial and fungal infections.

Composition

The immunomodulator Roncoleukin is a dosage form of recombinant human interleukin-2 (rIL-2) isolated and purified from Saccharomyces cerevisiae yeast cells, solubilizer-dodecyl sulfate sodium (DSN), stabilizer-D-mannitol and reducing agent-dithiothreitol (DTT).

Directions for use

Subcutaneously (p/k): dissolve the contents of the ampoule in 2-5 ml of 0.9% sodium chloride solution or water for injection.

Intravenously (IV): transfer the contents of the ampoule with Roncoleukin into 100 – 400 ml of 0.9% sodium chloride solution for injection.

Intranasally: dissolve the contents of the ampoule in 4 ml of 0.9% sodium chloride solution or water for injection.

Push into the nose: Use an ampoule of 100,000 IU for animals weighing up to 5 kg, an ampoule of 250,000 IU for animals weighing up to 20 kg and 500,000 IU for animals weighing more than 20 kg. Put two drops in each nostril at least 3-5 times a day.

Externally: dissolve the contents of the ampoule in 5-10 ml of 0.9% sodium chloride solution or water for injection and use as applications or surface irrigation.

Intercystic (intravesical): dissolve the contents of an ampoule of 100,000 to 250,000 IU in 5-50 ml (depending on the size of the animal) of 0.9% sodium chloride solution and inject through a catheter.

Orally: dissolve the contents of the ampoule in 2-10 ml of 0.9% sodium chloride solution or water for injection (depending on the size of the animal) and feed the animal.

Airosol: Dilute the contents of the ampoule in the required amount of 0.9% sodium chloride solution or water for injection and spray on the device or directly into the oral cavity of the animal.

When diluting Roncoleukin one should not shake the ampoule strongly to avoid foaming. The drug can be administered for therapeutic and preventive purposes starting from newborn age.

Special Instructions

Roncoleukin is compatible with most other drugs, but during its use some rules should be observed: Roncoleukin should not be injected with any other drugs and vaccines in one syringe (except vaccination of birds); Roncoleukin should not be injected in solutions containing glucose, because the activity of the drug is reduced. Livestock products after using Roncoleukin immunomodulator may be used for food purposes without restrictions.

Contraindications

Cattle and Horses with severe lesions of cardiac conduction system and valve apparatus are used with caution, in portions. Myocardial infarction in Cattle and Horses is a contraindication for using Roncoleukin.  Roncoleukin is not recommended to be administered to horses shortly before or immediately after exercise and also in hot weather due to hyperthermia effect. In malignant diseases of blood and lymph, Roncoleukin should be used with caution. It is not recommended to use Roncoleukin simultaneously with corticosteroid drugs for local or systemic use.

Side effects

Subcutaneous injection of Roncoleukin may be accompanied by a brief, up to 1 minute, painful “burning” reaction. In order to reduce this effect, water for injection should be used to dilute the drug.
