Vinpocetine has a vasodilator, antihypoxic, anti-aggregation action. It inhibits phosphodiesterase and increases the content of cyclic adenosine monophosphate in cells, which leads to a decrease of calcium in the cytoplasm of smooth muscle cells and relaxation of myofibrils. It dilates brain vessels, increases blood flow, mainly in ischemic areas, improves the supply of oxygen to the brain.

It promotes glucose utilization and increases catecholamine levels in the central nervous system and stimulates the metabolism of noradrenaline and serotonin in brain tissues. It reduces platelet aggregation and blood viscosity, increases red blood cell deformability and normalizes venous outflow against decreased resistance of cerebral vessels. Systemic arterial pressure slightly decreases.

In old age and old age, the sensitivity of cerebral vessels to the relaxing action of Vinpocetine increases due to sensitization of the adenylate cyclic adenosine monophosphate system during aging.

Pharmacokinetics

It is rapidly absorbed from the gastrointestinal tract, bioavailability is about 60%. The maximum concentration in blood plasma is reached after 1 hour. It easily penetrates through histohematic barriers (including the blood-brain barrier). The elimination half-life is about 5 hours.

Ingestion: 5-10 mg three times a day.

Interaction

When Vinpocetine is used with heparin therapy, the risk of hemorrhagic complications increases.

Special Instructions

With caution, use orally in arrhythmia, angina pectoris, unstable BP and decreased vascular tone.

Contraindications

– marked cardiac rhythm disorders;
– CHD (severe course);
– acute stage of hemorrhagic stroke;
– pregnancy;
– lactation (breast-feeding);
– hypersensitivity to the drug.

Side effects

Cardiovascular system: decreased BP, tachycardia, extrasystole, delayed intraventricular conduction.

Overdose

There have been no reported cases of overdose of the drug Vinpocetine.

Treatment: gastric lavage, administration of activated charcoal; if necessary symptomatic therapy is carried out.

Pregnancy use

Vinpocetine is contraindicated in pregnancy and during lactation (breastfeeding).
