Pharmacodynamics

The spectrum of action is associated with the presence of gamma-aminobutyric acid in the structure. The mechanism of action is due to the direct effect of pantocalcin on GABA-receptor-channel complex. It has neurometabolic, neuroprotective and neurotrophic properties. Increases resistance of the brain to hypoxia and toxic substances, stimulates anabolic processes in neurons. It has anticonvulsant effect, reduces motor excitability with simultaneous ordering of behavior. Increases mental and physical performance.

It promotes normalization of gamma-aminobutyric acid in chronic alcohol intoxication and subsequent withdrawal of ethanol. It shows analgesic effect. It is able to inhibit acetylation reactions participating in mechanisms of inactivation of novocaine and sulfonamides; due to this it is possible to achieve prolongation of the action of the latter. It causes inhibition of pathologically increased bladder reflex and detrusor tone.

Pharmacokinetics

It is rapidly absorbed in the gastrointestinal tract. Time of reaching maximum concentration in blood plasma is 1 h. The highest concentrations are formed in the liver, kidneys, gastric wall, skin. It penetrates through blood-brain barrier.

It is not metabolized. It is excreted unchanged during 48 hours (67.5% of dose – in the urine, 28.5% – in the faeces).

Indications

Urinary disorders, Acquired dementia, Low performance, Epilepsy, Impaired cerebral circulation, Seizures, Neurosis, Concussion and other craniocerebral injuries, Psycho-emotional stress, Attention and memory disorders

• Cognitive disorders in organic brain lesions and neurotic disorders;
• Cognitive disorders in organic brain lesions and neurotic disordersin complex therapy of cerebrovascular insufficiency caused by atherosclerotic changes in brain vessels; senile dementia (initial form), residual organic brain lesions in elderly people;
• cerebral organic insufficiency in patients with schizophrenia (in combination with neuroleptics, antidepressants);
• extrapyramidal hyperkinesis in patients with hereditary diseases of the nervous system (including.ч. Huntington’s chorea, hepatocerebral dystrophy, Parkinson’s disease);
• consequences of neuroinfections and craniocerebral injuries (as part of complex therapy);
• correction and prevention (covert therapy in concurrent administration) of side effects of neuroleptics; extrapyramidal neuroleptic syndrome (hyperkinetic and akinetic);
• epilepsy with delayed mental processes (in combination with anticonvulsants);
• psychoemotional overload, decreased mental and physical performance; to improve concentration and memorization;
• lowering disorders: enuresis, diurnal urinary incontinence, pollakiuria, imperative urges;

Children:

• mental retardation (mental, speech, motor development delay or combinations thereof);
• children’s cerebral palsy;
• stuttering (predominantly clonic form);
• Epilepsy (as part of combined therapy with anticonvulsants, especially for polymorphic seizures and minor epileptic seizures).

Composition

1 tablet contains:
active ingredient:
hopanthenic acid – 250.00 mg or 500.00 mg;
excipients:
magnesium hydroxycarbonate, calcium stearate, talc, potato starch.

How to take, the dosage

Ingestion.

Take 15-30 minutes after a meal.

A single dose for adults is 0.5-1 g, for children – 0.25-0.5 g, daily dose – 1.5-3 g (for adults), 0.75-3 g (for children). Duration of treatment course – from 1 to 4 months, in some cases – up to 6 months. After 3 to 6 months it is possible to carry out the second course of treatment.

Children. Mental retardation – 0.5 g 4-6 times a day, every day for 3 months. Delayed speech development – 0.5 g 3-4 times a day for 2-3 months.

The neuroleptic syndrome (as a corrector of the side effects of neuroleptic drugs). Adults – 0.5-1 g 3 times a day; children – 0.25-0.5 g 3-4 times a day. Duration of treatment course is 1-3 months.

Epilepsy. Children – 0.25-0.5 g 3-4 times a day; adults – 0.5-1 g 3-4 times a day for a long time (up to 6 months).

Hyperkinesias (tics). Children – 0.25-0.5 g 3-6 times daily for 1-4 months; adults – 1.5-3 g daily for 1-5 months.

Disorders of urination. Adults – 0.5-1 g 2-3 times a day (daily dose – 2-3 g); children – 0.25-0.5 g (daily dose – 25-50 mg/kg). Duration of the course of treatment – from 2 weeks to 3 months (depends on the severity of the disorders and the therapeutic effect).

The consequences of neuroinfections and craniocerebral injuries. 0,25 g 3-4 times a day.

To restore performance under increased stress and asthenic conditions.0.25 g 3 times a day.

This dosage form is not recommended for children under 3 years of age.

Interaction

The action of gopantenic acid is enhanced in combination with glycine, xydiphone.

Prolongs the effect of barbiturates and enhances the effects of drugs that stimulate the CNS, anticonvulsants, the effect of local anesthetics (procaine).

Prevents side effects of the following drugs: phenobarbital, carbamazepine, antipsychotics (neuroleptics).

Synopsis

Round, white, flat-cylindrical tablets with a bevel for 500 mg dosage and a cross-shaped bevel for 250 mg dosage.

Contraindications

• Hypersensitivity;
• acute renal failure;
• pregnancy (1st trimester)

Side effects

Allergic reactions (rhinitis, conjunctivitis, skin rashes) are possible.

Overdose

Symptoms: Characterized by increased severity of side effects.

Treatment:Gastric lavage, administration of activated charcoal, symptomatic therapy.

Pregnancy use

It is contraindicated in the first trimester of pregnancy.
