Pharmacodynamics
The spectrum of action is associated with the presence of gamma-aminobutyric acid in the structure. The mechanism of action is due to the direct effect of pantocalcin on GABA-receptor-channel complex. It has neurometabolic, neuroprotective and neurotrophic properties. Increases resistance of the brain to hypoxia and toxic substances, stimulates anabolic processes in neurons. It has anticonvulsant effect, reduces motor excitability with simultaneous ordering of behavior. Increases mental and physical performance.
It promotes normalization of gamma-aminobutyric acid in chronic alcohol intoxication and subsequent withdrawal of ethanol. It shows analgesic effect. It is able to inhibit acetylation reactions participating in mechanisms of inactivation of novocaine and sulfonamides; due to this it is possible to achieve prolongation of the action of the latter. It causes inhibition of pathologically increased bladder reflex and detrusor tone.
Pharmacokinetics
It is rapidly absorbed in the gastrointestinal tract. Time of reaching maximum concentration in blood plasma is 1 h. The highest concentrations are formed in the liver, kidneys, gastric wall, skin. It penetrates through blood-brain barrier.
It is not metabolized. It is excreted unchanged during 48 hours (67.5% of dose – in the urine, 28.5% – in the faeces).
Indications
Cognitive impairment in organic brain lesions and neurotic disorders;
as part of complex therapy for cerebrovascular insufficiency caused by atherosclerotic changes in cerebral vessels; senile dementia (initial form), residual organic brain damage in mature and elderly people;
cerebral organic failure in patients with schizophrenia (in combination with antipsychotics, antidepressants);
extrapyramidal hyperkinesis in patients with hereditary diseases of the nervous system (including Huntington’s chorea, hepatocerebral dystrophy, Parkinson’s disease);
consequences of neuroinfections and traumatic brain injuries (as part of complex therapy);
correction and prevention (cover-up therapy with simultaneous prescription) of side effects of antipsychotics; extrapyramidal neuroleptic syndrome (hyperkinetic and akinetic);
epilepsy with slowing of mental processes (in combination with anticonvulsants);
psycho-emotional overload, decreased mental and physical performance; to improve concentration and memory;
urinary disorders: enuresis, daytime urinary incontinence, pollakiuria, urgency;
Children:
mental retardation (delayed mental, speech, motor development or a combination thereof);
cerebral palsy;
stuttering (mainly clonic form);
epilepsy (as part of combination therapy with anticonvulsants, especially for polymorphic seizures and petit mal seizures).

Composition
1 tablet contains:

hopantenic acid – 250.00 mg or 500.00 mg;
excipients:
magnesium hydroxycarbonate, calcium stearate, talc, potato starch.
Pregnancy
Contraindicated in the first trimester of pregnancy.
Contraindications
Hypersensitivity;
acute renal failure;
pregnancy (1st trimester).
Side Effects
Allergic reactions are possible (rhinitis, conjunctivitis, skin rashes).
Interaction
The effect of hopantenic acid is enhanced in combination with glycine and xydiphone.
Prolongs the effect of barbiturates; enhances the effects of drugs that stimulate the central nervous system, anticonvulsants, and the effect of local anesthetics (procaine).
Prevents the side effects of the following drugs: phenobarbital, carbamazepine, antipsychotics (neuroleptics).
Overdose
Symptoms: characterized by increased severity of side effects.
Treatment: gastric lavage, administration of activated carbon, symptomatic therapy.

In a place protected from light at a temperature not exceeding 25°C.
