Pharmacodynamics
Simethicone is a chemically inert methylsiloxane polymer containing about 5% silicon dioxide.
Simethicone has foaming and anti-foaming properties and significantly reduces the surface tension of purified water and model surfactant solutions. The results of foaming experiments demonstrated complete foam destruction within 3-6 seconds when adding a liquid suspension of simethicone at a concentration of 0.1 mg/ml. Below this threshold, complete foam destruction does not occur. As a defoamer simethicone is also effective at a concentration of 0.1 mg/ml. This effect lasts for more than 24 hours.
The mechanism of action is based on the distribution of the silicone liquid over the surface of the foam film as well as on a local change in surface tension in the contact area. This causes rapid drainage of the liquid, which helps to break up the film, with the consequent possibility for the released gas to be absorbed or expelled naturally.

Pregnancy
There are no clinical data on the effects of simethicone when used during pregnancy.
Simethicone can be used during pregnancy only if the expected benefit of therapy for the mother outweighs the potential risk to the fetus, taking into account the fact that simethicone is not physiologically active and is not absorbed from the gastrointestinal tract. There is no data on the excretion of simethicone in breast milk.
