Pharmacological properties
Pharmacodynamics
Magnesium drug. It is the most important macronutrient, magnesium performs more than 300 enzymatic reactions, regulation of cell permeability and neuromuscular excitability. Necessary for many energy processes, is involved in the metabolism of proteins, fats, carbohydrates and nucleic acids. Magnesium has a curare-like effect on the terminations of cholinergic neurons, with a decrease in acetylcholine release; magnesium and calcium interact partly synergistically in the body; however, while competitively inhibiting calcium, magnesium can also affect its binding sites as a “physiological calcium antagonist”.
Pharmacokinetics
The absorption is approximately 35-40% of the administered dose. Hypomagnesemia stimulates absorption of magnesium ions. The presence of orotic acid salts promotes magnesium absorption.
Magnesium is excreted by the kidneys, excretion decreases in magnesium deficiency and increases in magnesium excess.

Composition per tablet:
Active ingredient: magnesium orotate dihydrate – 500 mg;
Excipients: colloidal silica, croscarmellose sodium, microcrystalline cellulose, corn starch, povidone-K30, lactose monohydrate, sodium cyclamate, talc, magnesium stearate.
How to take, the dosage
Interaction
The simultaneous administration of magnesium-containing drugs with preparations of iron, sodium fluoride and tetracyclines may reduce the absorption of the latter. In addition, aminoquinolines, quinidine, penicillamine should not be administered simultaneously with magnesium preparations. Therefore, these drugs should be taken 2-3 hours apart. Oral contraceptives, diuretics, muscle relaxants, glucocorticosteroids, insulin reduce the effect of the drug.
Magnesium should be used with caution if you are taking magnesium-containing medications such as antacids or laxatives, or potassium-saving diuretics or calcium-containing medications. When used concomitantly with drugs containing aluminum, aluminum absorption in the body may be increased.
Magnesium should be used with caution when used concomitantly with foxglove drugs because concomitant use may interfere with absorption of foxglove drugs.
Some antibiotics (aminoglycosides), cisplatin and cyclosporine A may lead to increased magnesium excretion.
Some diuretics (thiazides and furosemide), cetuximab and erlotinib, proton pump inhibitors (omeprazole and pantoprazole), foscarnet, pentamidine, rapamicin and amphotericin B may cause magnesium deficiency and thus lead to reduced effect of Magnerot®.
Special Instructions
The drug contains lactose monohydrate, therefore its use is not recommended in patients with hereditary galactosemia, glucose and galactose malabsorption syndrome or lactase deficiency.
