Pimobendan is a benzimidazole-pyridazinone derivative and has positive inotropic and vasodilatory (vasodilator) effects. Unlike cardiac glycosides positive inotropic effect of pimobendan is caused by two mechanisms: increase of cardiac muscle fibers sensitivity to calcium and inhibition of phosphodiesterase III (vasodilating effect).
Due to positive inotropic and vasodilatory effects in heart failure the drug increases strength of heart contractions and decreases both preload and postload.
During intravenous administration of the drug the bioavailability of pimobendanum is 100%. After intravenous administration its distribution volume is 2.6 L/kg, indicating that pimobendan is rapidly distributed in tissues. Binding to plasma proteins is 93%.
The half-life of pimobendan in blood plasma is 0.4±0.1 hours, and the elimination half-life of its active metabolite is 2.0±0.3 hours. Almost all amount of pimobendan is eliminated from animals via feces.
Vetmedin by degree of exposure on the body is classified as a low-hazard substance (4 class of hazard according to GOST 12.1.007-76), it has no sensitizing, embryotoxic, teratogenic and mutagenic properties.
Indications
Vetmedin is used in dogs to treat heart failure caused by dilated cardiomyopathy (DCMP), bicuspid or tricuspid valve failure.
Composition
1 ml of the drug contains 0.75 mg of pimobendan as an active ingredient, as well as excipients: hydroxypropyl-13-cyclodextrin, sodium dodecahydrate hydrophosphate, sodium dihydrophosphate dihydrate, sodium hydroxide, hydrochloric acid, water for injection.
How to take, the dosage
Vetmedin for injection is administered to animals once intravenously at a dose of 0.15 mg pimobendan per 1 kg of the dog’s body weight (i.e. 2 ml/10 kg of body weight) to stop an acute attack of heart failure.
Contraindications
Contraindication to the use of Vetmedin for injection is hypersensitivity of the animal to its components.
The use of Vetmedin is prohibited in hypertrophic cardiomyopathy and clinical conditions in which the cardiac output cannot be increased due to functional or anatomical features (eg, aortic stenosis).
Simultaneous use of the drug Vetmedin with slow calcium channel blocker (verapamil) and beta-blocker (propranololol) weakens the effect of pimobendan. The drug can be used simultaneously with the cardiac glycoside ouabain. There is no information on peculiarities of interaction with drugs of other pharmacological groups.
The drug is not intended for use in productive animals.
