Thioridazine, the active ingredient in Thioril, is a piperidine derivative of phenothiazine. It has antipsychotic, tranquilizing, antidepressant, antipruritic, alpha-adrenoblocking and m-cholinoblocking effects.

In small doses the substance reduces anxiety and tension, has an anxiolytic effect, in large doses it shows neuroleptic properties. When administered in medium therapeutic doses, it does not cause somnolence and extrapyramidal disorders. Besides, thioridazine has weak antiemetic and moderate hypotensive effects. The central antiemetic effect of the drug is explained by inhibition or blockade of dopamine Og-receptors in the chemoreceptor trigger zone of the cerebellum, peripheral – by blockade of the vagus nerve in the gastrointestinal tract (GIT).

The antipsychotic (neuroleptic) action of Tioril is explained by blockade of postsynaptic dopaminergic receptors in the mesolimbic structures of the brain. Thioridazine inhibits release of hypothalamic and pituitary hormones (but pituitary prolactin release increases due to blockade of dopamine receptors), has alpha-adrenoblocking action.

With caution use in patients with cardiac rhythm disturbances, heart disease, severe respiratory diseases, renal failure, Parkinson’s disease, closed-angle glaucoma, prostatic hypertrophy, myasthenia, epilepsy, pheochromocytoma.

At the time of treatment it is not allowed to drink alcohol.
Patients with liver disease require regular monitoring of liver function. Patients who are elderly, emaciated, or have kidney or liver disease are advised to prescribe a low starting dose with gradual increase in dose.

Impact on the ability to drive and operate machinery:

During treatment, refrain from potentially hazardous activities requiring increased attention and rapid psychomotor reactions.
