Pharmacodynamics
Fenotropil is a nootropic medicine with evident anti amnesic effect, direct activation of cerebral integrative activity, contributes to memory consolidation, improves concentration and mental performance, facilitates learning process, increases information transfer speed between cerebral hemispheres, increases brain tissue resistance to hypoxia and toxic effects, has anticonvulsant and anxiolytic effects, regulates activation and inhibition processes.
Phenotropil has a positive effect on metabolic processes and blood circulation in the brain, stimulates redox processes, increases the body’s energy potential by utilization of glucose, improves regional blood flow in ischemic brain areas. It increases norepinephrine, dopamine and serotonin content in the brain, does not affect the level of GABA content, binds neither to GABA nor GABA receptors and has no significant effect on spontaneous bioelectrical activity of the brain.
Phenotropil has no effect on respiration and cardiovascular system, shows slight diuretic effect, has anorexigenic activity when used regularly.
The stimulating effect of FENOTROPIL is seen in its ability to produce a moderate effect on motor reactions, increased physical performance, pronounced antagonism to the cataleptic action of neuroleptics and weakening of the pronounced hypnotic effect of ethanol and hexenal.
The psychostimulant effect of PENOTROPIL predominates in the ideational sphere.
Moderate psychostimulant effect of the drug is combined with anxiolytic activity, improves mood, has some analgesic effect, increasing the threshold of pain sensitivity.
The adaptogenic effect of FENOTROPIL is shown in increasing the resistance of the organism to stress under conditions of excessive mental and physical strain, fatigue, hypokinesia and immobilization, low temperatures.
Phenotropil causes improvement of vision, which is manifested by increase of visual acuity, brightness and visual fields.
Phenotropil improves blood flow to the lower extremities.
Phenotropil stimulates the production of antibodies in response to antigen introduction, indicating that it has immunostimulatory properties, but at the same time it does not promote immediate hypersensitivity and does not change the allergic skin inflammatory response following the introduction of a foreign protein.
Phenotropyl does not cause drug dependence, tolerance or withdrawal symptoms.
The action of FENOTROPIL starts with a single dose, which is important when using the drug in extreme conditions.
PHENOTROPIL has no teratogenic, mutagenic, carcinogenic and embryotoxic properties. Toxicity is low, the lethal dose in an acute experiment is 800 mg/kg.
Pharmacokinetics
Phenotropyl is rapidly absorbed, penetrates various organs and tissues, easily passes through the blood-brain barrier. Absolute bioavailability of the drug when administered orally is 100%. Maximum concentration in blood is reached after 1 hour, half-life is 3-5 hours. Fenotropil is not metabolized in the body and is excreted unchanged. About 40% of the drug is excreted with urine and 60% of the drug is excreted with bile and sweat.
Indications
Diseases of the central nervous system of different genesis, especially associated with vascular diseases and disorders of metabolic processes in the brain, intoxication (in particular in post-traumatic states and phenomena of chronic cerebrovascular insufficiency), accompanied by deterioration of intellectual and mental functions, decreased motor activity.
Neurotic conditions, manifested by lethargy, increased exhaustion, decreased psychomotor activity, impaired attention, and memory impairment.
Disorders of learning processes.
Depressions of mild to moderate severity.
Psychorganic syndromes manifested by intellectual and mental disturbances and apathetic-abulatory phenomena, as well as lethargic-apathetic states in schizophrenia.
Convulsive states.
Obesity (of alimentary and constitutional genesis).
Preventing hypoxia, increasing resistance to stress, correction of the functional state of the body in extreme conditions of professional activity in order to prevent the development of fatigue and increase mental and physical performance, correction of the daily biorhythm, the inversion of the sleep-wake cycle;
Chronic alcoholism (to reduce the phenomena of asthenia, depression, intellectual and mental disorders).

How to take, the dosage
Phenotropil is used orally, immediately after meals. The dose of the drug and duration of treatment should be determined by the doctor. Doses vary depending on the characteristics of the patient’s condition. The average single dose is 150 mg (100 mg to 250 mg); the average daily dose is 250 mg (200 mg to 300 mg). The maximum tolerated dose is 750 mg per day. It is recommended to divide the daily dose into 2 doses. The daily dose up to 100 mg should be taken once in the morning, and the daily dose over 100 mg should be divided into two doses. The duration of treatment may vary from 2 weeks to 3 months. The average duration of treatment is 30 days. If necessary, the course can be repeated in a month.
To improve performance, 100-200 mg once in the morning hours, for 2 weeks (for athletes, 3 days).
The recommended duration of treatment for patients with alimentary-constitutional obesity is 30-60 days at a dose of 100-200 mg once a day (in the morning hours). It is not recommended to take FENOTROPIL later than 3 pm.
Interaction
Phenotropil may increase the effect of drugs that stimulate the central nervous system, antidepressants and nootropics.
Special Instructions
In excessive psycho-emotional exhaustion on the background of chronic stress and fatigue, chronic insomnia, a single administration of FENOTROPIL in the first day may cause a sharp need for sleep. In such patients on outpatient basis it should be recommended to start course administration of the drug on days off.

Fenotropil is used with caution in patients with severe organic lesions of the liver and kidneys, severe arterial hypertension, patients with atherosclerosis, also in patients who have previously suffered from panic attacks, acute psychotic states with psychomotor agitation – due to the possibility of worsening of anxiety, panic, hallucinations and delirium, as well as in patients who are prone to allergic reactions to nootropic drugs of pyrrolidone group.

Pregnancy use
FENOTROPIL should not be prescribed during pregnancy and lactation due to lack of clinical trial data.
Pediatric use
The use of FENOTROPIL in children is not recommended due to the lack of data on the use of the drug in children.
