Pensital is amylolytic, lipolytic, proteolytic, making up for the deficiency of pancreatic enzymes.

Pharmacodynamics

Pensital compensates insufficiency of external secretory function of the pancreas, has proteolytic, amylolytic and lipolytic action.

Pancreatin contains enzymes (lipase, alpha-amylase, trypsin, chymotrypsin) which aid in the breakdown of proteins into amino acids, fats into glycerol and fatty acids, starch into dextrins and monosaccharides. It improves the functional state of the gastrointestinal tract and normalizes digestion.

Trypsin inhibits stimulated pancreatic secretion with analgesic effect.

Pancreatic enzymes are released from the dosage form in the alkaline environment of the small intestine because they are protected from the action of gastric juice by the coating.

The maximum enzymatic activity is observed 30-45 minutes after oral administration.

Each tablet contains:

Pancreatin 212.5 mg with enzyme activity:
lipase 6000 units FIP
amylase 4500 units FIP
protease 300 units FIP

How to take, the dosage

Overly, during or after meals, without chewing and with non-alkaline liquids (water, fruit juices).

The dose is determined individually, depending on the degree of digestive disorders.

Adults are usually prescribed 1-2 tablets 3 times a day. In children the drug is used as prescribed by the doctor.

The duration of treatment may vary from a few days (when there is a digestive disturbance due to errors in the diet) to a few months or years (if permanent replacement therapy is necessary).

Interaction

Concomitant use of pancreatin with iron preparations may reduce the absorption of the latter.

Concomitant use of antacids containing calcium carbonate and/or magnesium hydroxide may lead to decreased effectiveness of pancreatin.

Special Instructions

In cystic fibrosis, high doses of pancreatin are not recommended because of the increased risk of strictures (fibrous colonopathy).

The dose should be adequate to the amount of enzymes required for fat absorption, taking into account the quality and quantity of food consumed.

In long-term use, iron preparations are prescribed at the same time.

Contraindications

Side effects

Allergic reactions. In some cases – diarrhea, constipation, feeling of discomfort in the stomach, nausea (causal relation of development of these reactions with the action of pancreatin is not established, because these phenomena refer to the symptoms of external excretory pancreatic failure).

In long-term use in high doses development of hyperuricosuria, increase of plasma uric acid level is possible. In cystic fibrosis in case of excessive doses of pancreatin it is possible to develop strictures (fibrous colonopathy) in the ileocecal region of the ascending colon.

When pancreatin is used in high doses in children perianal irritation and irritation of the oral mucosa may occur.

Overdose

Symptoms: hyperuricosuria, hyperuricemia. In children – constipation.

Treatment: drug withdrawal, symptomatic therapy.

Pregnancy use

The safety of pancreatin use in pregnancy has not been adequately studied.

The use is possible in cases when the expected benefit to the mother exceeds the potential risk to the fetus.
