Pharmacological action – myotropic antispasmodic, vasodilator.

Pharmacodynamics

Benciclane is a myotropic antispasmodic with pronounced vasodilatory action. The vasodilatory effect of benziclane is mainly due to its ability to block calcium channels, antiserotonin action and, to a lesser extent, blockade of sympathetic ganglia.

In addition, benziclane has an antispasmodic effect on the visceral musculature (gastrointestinal, genitourinary, respiratory organs). The drug causes a slight increase in heart rate. Its mild tranquilizing effect is also known.

Benciclane may cause dose-dependent inhibition of Na⁺/K⁺-dependent ATPase and platelet and erythrocyte aggregation, as well as increased erythrocyte elasticity. These effects are observed mainly in peripheral vessels, coronary arteries and cerebral vessels.

Indications

Vascular diseases

Diseases of peripheral vessels: Raynaud’s disease, other diseases with acrocyanosis and vasospasm, as well as chronic obliterating arterial diseases.
Diseases of cerebral vessels: in the complex therapy of acute and chronic cerebral ischemia.

Removes spasm of the internal organs

Gastrointestinal diseases: gastroenteritis of various etiology (especially infectious), infectious and inflammatory colitis, functional diseases of the large intestine, tenesmus, postoperative flatulence, cholecystitis, gallstone disease, condition after cholecystectomy, motility disorders in dyskinesia of the sphincter of the Odzi, stomach or duodenal ulcer – in combination with other medications.

Urological syndromes: urinary bladder spasms and tenesms, concomitant therapy of urolithiasis (in combination with analgesics for renal colic), preparation for instrumental methods of investigation.

Active ingredient

Composition

Each 2 ml ampule contains 50 mg of the active ingredient bencyclan fumarate, as well as excipients: sodium chloride 8 mg for parenteral dosage forms and water for injections up to 2 ml.

How to take, the dosage

Solution for intravenous injections (after dilution) or infusion.

Vascular diseases
Infusion:
In vascular diseases the drug can be used in a daily dose of 200 mg divided into 2 infusions. 100 mg (4 ml) of the drug is diluted in 100 – 200 ml of isotonic NaCl solution. The drug is administered by intravenous drip for 1 hour 2 times a day.

Removes spasm of internal organs
Injections:
In acute cases the drug is administered into a vein slowly 2-4 ampoules (4-8 ml) diluted with saline solution to 10 – 20 ml or intramuscularly 2 ml.
Treatment course of 2-3 weeks, if necessary, followed by transferring the patient to the drug Galidor tablets.

Special Instructions

Simultaneous use of Galidor with sedatives leads to increased drowsiness.

Contraindications

Side effects

Overdose

Symptoms: increased heart rate, decreased blood pressure, predisposition to collapse, urinary incontinence, drowsiness, restlessness, and in severe cases, epileptiform seizures. Significant overdose may cause tonic and clonic seizures.

Treatment: No specific antidote is known. In case of overdose symptomatic treatment should be used. Benzodiazepines are recommended for treatment of seizures. There is no data on possible excretion of benzicline by dialysis.

Pregnancy use

The data from preclinical studies showed no embryotoxic or teratogenic effects. However, sufficient reliable studies on the use of the drug during pregnancy and lactation in humans have not been conducted. Therefore, administration of the drug to patients in the first trimester of pregnancy is not recommended. During lactation the drug should be refrained from prescribing, or consider stopping breastfeeding during treatment.
