Clotrimazole is an imidazole derivative, a broad-spectrum antifungal agent. Antifungal effect is associated with disruption of the synthesis of ergosterol, which is part of the cell membrane of fungi, which causes changes in its structure and properties and leads to cell lysis.
In low concentrations it is fungistatic, in high concentrations it is fungicidal, and not only on proliferating cells. In fungicidal concentrations, it interacts with mitochondrial and peroxidase enzymes, resulting in an increase in hydrogen peroxide concentration to toxic levels, which also contributes to the destruction of fungal cells.
It is effective against dermatophytes (Epidermophyton, Microsporum, Trichophyton), yeasts (mainly Candida albicans), mold fungi and protozoa, as well as the causative agent of variegated lichen Pityriasis versicolor and erythrazma. It has antimicrobial action against Gram-positive microorganisms and anaerobes. Clotrimazole has no effect on lactobacilli.
When applied topically there is little adsorption of clotrimazole from mucous membranes.
