Tereflex stimulates the regeneration of cartilage tissue. Glucosamine and chondroitin sulfate sodium are involved in the synthesis of connective tissue, helping to prevent the processes of cartilage destruction and stimulate tissue regeneration.
The administration of exogenous glucosamine increases the production of the cartilage matrix and provides nonspecific protection against cartilage damage. Other possible effects of glucosamine are protection of damaged cartilage from metabolic damage caused by NSAIDs and GCS and its own moderate anti-inflammatory effect.
Chondroitin sulfate serves as an additional substrate for the formation of a healthy cartilage matrix. It stimulates the formation of hyaluronone, synthesis of proteoglycans and type II collagen and also protects hyaluronone from enzymatic degradation (by inhibiting the activity of hyaluronidase), maintains viscosity of synovial fluid, stimulates mechanisms of cartilage repair and inhibits the activity of those enzymes (elastase and hyaluronidase) which degrade cartilage.
In the treatment of osteoarthritis, it relieves the symptoms of the disease and reduces the need for NSAIDs.
Pharmacokinetics
Glucosamine
Absorption. The bioavailability of glucosamine when taken orally is 25% (effect of first passage through the liver).
Metabolism. After absorption of oral dose radioactively labeled glucosamine is first detected in plasma and later penetrates into tissues. The highest concentrations are found in the liver, kidneys and articular cartilage. About 30% of the dose taken persists in bone and muscle tissues for a long time.
Elimination. It is excreted mainly unchanged by the kidneys; partially – through the intestine. T1/2 of the preparation is 68 hours.
Chondroitin sulfate
Absorption. When chondroitin sulfate is taken orally once in dose of 0.8 g (or 2 times per day in dose of 0.4 g) the concentration in plasma increases within 24 h. Absolute bioavailability is 12%.
Metabolism. It is metabolized by desulphurization.
Elimination. It is excreted by the kidneys. T1/2 is 310 min.
