A herbal laxative (senna or alexandria leaf, or cassia spicifolia leaf).
The pharmacological action is due to the anthraglycosides (mainly senosides A and B), but also to other glycosides. When used orally, senna glycosides pass through the stomach and small intestine unchanged without being absorbed.
In the large intestine, anthraglycosides are broken down by intestinal bacteria into pharmacologically active anthrons and anthranols, which irritate colonic interreceptors, increase peristalsis and accelerate bowel emptying.
Antrons and anthranols prevent absorption of water and electrolytes and promote diffusion of water into the intestinal lumen. Due to the osmotic effect, the stool masses are loosened and their volume increases, contributing to the laxative effect of anthraglycosides.
The laxative effect occurs after 6 – 10 hours. If the drug is used for a long time, the intensity of its effect may decrease due to decrease of potassium content in the body.
The drug is well tolerated by patients; due to the low content of resinous substances the intestines are not irritated.

How to take, the dosage
Senadexin is taken before bedtime.
Adults and children over 12 years: ½-1 cc of the lozenge or 1-2 tablets. If there is no effect, the dosage of the drug is increased to 3 tablets per dose (a few days after the first dose).
Children 1-6 years are prescribed 1/3 of a tablet. For children 6-12 years old, half the adult dosage.
Interaction
In long-term use or use in high doses the drug increases the effect of cardiac glycosides and antiarrhythmic drugs due to decrease of potassium in the body impairs absorption of tetracyclines.
In concomitant use with thiazide diuretics, adrenal hormones, licorice root preparations, the risk of hypokalemia increases.
Decreases the effectiveness of drugs that are slowly absorbed in the gastrointestinal tract.
Special Instructions
Long-term use of Senadexin leads to dehydration, diarrhea with metabolic syndrome, atony of the colon, digestive disorders.
In some cases it reduces the speed of psychomotor reactions (depending on individual characteristics).

Side effects
Senadexin administration may lead to hematuria, proteinuria, hypokalemia, intestinal melanosis, flatulence, discolored urine, dyspeptic disorders, skin rash, fatigue, seizures, vascular collapse, electrolyte-water imbalance, and confusion.
Overdose
Symptoms: in case of overdose diarrhea may develop, leading to disorders of water-electrolyte balance.
Treatment: symptomatic. The use of the drug should be cancelled.
Pregnancy use
The use of the drug in pregnant women is associated with a higher probability of side effects (abdominal pain, diarrhea).
Breast-feeding should be stopped during treatment with the drug, because the drug components may penetrate into the breast milk and cause frequent liquid stools in the infant.
