The antipsychotic action of Chlorprotixen is associated with its blocking effect on dopamine receptors. Blockade of these receptors is also associated with antiemetic and analgesic properties of the drug.
Chloroprotixen is able to block 5-NT2 receptors, α1-adrenoreceptors and H1-histamine receptors, which determines its adrenoblocking hypotensive and antihistamine properties.
Pharmacokinetics
The oral bioavailability of chlorprotixen is approximately 12%. Chlorprotixen is rapidly absorbed from the intestine, Cmax in serum is reached after 2 hours. T1/2 is about 16 hours.
Chlorprotixene penetrates the placental barrier and is excreted in small amounts with breast milk.
The metabolites have no neuroleptic activity and are excreted in the feces and urine.

How to take, the dosage
Psychosis, including schizophrenia and manic states.
Treatment begins with 50-100 mg/day, gradually increasing the dose until optimal effect is achieved, usually to 300 mg/day. In individual cases, the dose may be increased to 1200 mg/day. The maintenance dose is usually 100-200 mg/day. The daily dose of Chloroprotixen Zentiva is usually divided into 2-3 doses; given the pronounced sedative effect of Chloroprotixen Zentiva, it is recommended that a smaller portion of the daily dose be administered during the day and a larger portion in the evening.
Hangover withdrawal syndrome in alcoholism and drug addiction.
The daily dose, divided into 2-3 doses, is 500 mg. The course of treatment usually lasts 7 days. After withdrawal symptoms disappear, the dose is gradually reduced. The maintenance dose of 15-45 mg/day helps to stabilize the condition, reducing the risk of another binge.
In elderly patients with hyperactivity, irritability, agitation, confusion, 15-90 mg/day is prescribed. The daily dose is usually divided into 3 doses.
In children for correction of behavioral disorders, Chlorprotixen Zentiva is prescribed at the rate of 0.5-2 mg/kg body weight.
Depressive states, neuroses, psychosomatic disorders.
Chloroprotixen Zentiva may be used for depression, especially combined with anxiety, tension, as an adjunct to antidepressant therapy or alone. Interaction
The CNS depressant effect of chlorprotixene may be enhanced with concomitant use of ethanol and ethanol-containing drugs, anesthetics, opioid analgesics, sedatives, hypnotics, neuroleptics.
Anticholinergic effect of chlorprotixene is enhanced with concomitant use of anticholinergic, antihistamine and antiparkinsonian drugs.
The drug increases the effect of hypotensive drugs.
The simultaneous use of chlorprotixen and adrenaline may lead to arterial hypotension and tachycardia.
The use of chlorprotixen leads to a decrease in the seizure threshold, which requires additional adjustment of the dose of antiepileptic drugs in patients with epilepsy.
Chlorprotixen’s ability to block dopamine receptors reduces the effectiveness of levodopa.
Extrapyramid disorders may occur with concomitant use of phenothiazines, metoclopramide, haloperidol, reserpine.
Special Instructions
Chloroprotixen should be administered with caution in patients with epilepsy, parkinsonism, with pronounced atherosclerosis of cerebral vessels, with a tendency to collapse, with severe cardiovascular and respiratory failure, with severe liver and renal function disorders, diabetes, prostatic hypertrophy.
The use of Chlorprotixen may cause false positive result in urine immunobiological test for pregnancy, false increase in bilirubin level in blood, changes in QT interval on electrocardiogram.
During the drug treatment it is recommended to refrain from drinking alcohol, avoid excessive insolation.
Impact on the ability to drive and operate machinery
