Metadoxil, tablets 500 mg 30 pcs

Metadoxil is a detoxifying, hepatoprotective, anti-alcoholic drug.

Pharmacodynamics

Hepatoprotective effect of the active substance is due to membrane stabilizing effect and is based on the ability to restore the ratio of saturated and unsaturated free fatty acids. As a result of this action the resistance of hepatocytes to the action of lipid peroxidation which occurs under the influence of various toxic agents increases.

The detoxifying effect of methadoxine is due to the activation of liver enzymes involved in the metabolism of ethanol – alcohol dehydrogenase and acetaldehyde dehydrogenase, which accelerates the removal of ethanol and acetaldehyde from the body and thus reduces their toxic effects.

Metadoxin prevents accumulation of triglycerides in hepatocytes and also prevents formation of fibronectin and collagen, which significantly slows down the process of cirrhosis formation.

The drug reduces mental and somatic manifestations of hangover syndrome, reduces the time of withdrawal syndrome relief.

Metadoxine activates cholinergic and GABAergic neurotransmitter systems, improves thinking and short-term memory functions, prevents motor agitation caused by ethanol.

Metadoxine has nonspecific antidepressant and anxiolytic effects, reduces craving for alcohol.

Pharmacokinetics

It is rapidly absorbed from the gastrointestinal tract and has high bioavailability (60-80%). Binding to plasma proteins is 50%. It is metabolized in the liver to active metabolites – pyridine and pyrrolidone carboxylate. T_1/2 in per oral and parenteral administration is 40-60 min. 45-50% of the drug is excreted in the urine within 24 hours, 35-50% of the drug is excreted in the feces within 96 hours.

How to take, the dosage

Acute alcohol intoxication: 300-600 mg (5-10 ml) in m/m or 300-900 mg (5-15 ml) in v/v, depending on the degree of intoxication.

In case of intravenous administration the required dose of the drug is diluted in 500 ml of isotonic sodium chloride solution or 5% dextrose (glucose) solution and administered by drip for 1.5 hours.

The complex therapy of liver diseases and chronic alcoholism: orally: 1 tablet 2 times a day (1000 mg) 15-30 minutes before a meal, the dose can be increased to 3 tablets (1500 mg) a day. The course of therapy should be at least 90 days.

Alcohol withdrawal syndrome: 900 mg (15 ml) as an IV infusion (see above) once a day for 3-7 days depending on the patient’s condition.

Interaction

When used together, it reduces the effectiveness of levodopa.

Special Instructions

With caution, the drug should be prescribed in Parkinson’s disease if levodopa is used at the same time, because methadoxine reduces the effectiveness of the latter.

Contraindications

The ampoules of the solution for injection contain sodium metabisulfite. This substance can cause allergic reactions or worsen asthma attacks in predisposed people, especially those with bronchial asthma.

Side effects

Allergic reactions. In patients with bronchial asthma, bronchospasm may develop when using the solution for injection.

Overdose

There are currently no reported cases of overdose of Metadoxil.

Pregnancy use

Metadoxil is contraindicated in pregnancy and lactation.
