Pharmacological properties

Pharmacodynamics

Linex® Forte normalizes intestinal microflora. One capsule of Linex® Forte contains lactic acid bacteria (Lactobacillus acidophilus (LA-5) and Bifidobacterium animalis subsp. lactis (BB-12)), each capsule contains 109 colony forming units of live lyophilized lactic acid bacteria. Live lactic acid bacteria are a normal component of the natural intestinal microflora. The main functions of lactic acid bacteria:

• protective: synthesis of substances with antibacterial activity
• enzyme-producing: hydrolysis of fiber, proteins, fats, starch, deconjugation of bile acids
• synthetic: synthesis of vitamins B, C, K, amino acids, organic acids, etc.д. Lactic acid bacteria LA-5 and BB-12 inhibit the growth of pathogenic bacteria by:
• lowering the pH in the gut (a result of the ability of LA-5 to synthesize lactic acid and BB-12 to synthesize acetic and succinic acids in addition to lactic acid);
• producing metabolites that have toxic effects against pathogenic bacteria (H2O2 formation);
• competes with pathogenic bacteria for nutrients;
• interacting with adhesion receptors, thus preventing colonization by other, potentially pathogenic microorganisms.

Pharmacokinetics

Lactic acid bacteria, such as LA-5 and BB-12, act directly in the gastrointestinal tract and the drug is not systemically absorbed when ingested. Standard pharmacokinetic studies are thus not applicable. The resistance of LA-5 and BB- 12 to gastric juice and bile allows the strains to survive in large numbers as they pass through the stomach and 12 duodenum. Both strains attach to the intestinal mucosa. Like other microorganisms, they are gradually eliminated from the gastrointestinal tract by peristalsis and defecation.

Indications

Treatment and prevention of intestinal dysbacteriosis. Intestinal dysbacteriosis has the following symptoms: diarrhea (diarrhea), dyspepsia (digestive disorders), constipation, flatulence (bloating), nausea, belching, vomiting, abdominal pain, allergic skin reactions are possible.

Active ingredient

Composition

1 capsule contains: Active ingredient: Probio-tec® AB Blend 64 powder (Probio-tec® AB Blend 64) – 60,000 mg (the powder contains the active ingredients: Lactobacillus acidophilus (LA-5) – 13.8 mg and Bifidobacterium animalis subsp. lactis (BB-12) – 4.2 mg; excipients: anhydrous dextrose – 29.1 mg, microcrystalline cellulose – 6 mg, potato starch 6 – mg, magnesium stearate – 0.9 mg). Excipients: Beneo Synergy 1 (Beneo® Synergy 1) – 171.500 mg (powder contains 90-94% inulin and 6-10% oligofructose (dextrose + fructose + sucrose)); magnesium stearate – 3.500 mg. Capsule body composition: hypromellose – 33.149 mg, titanium dioxide (E 171) – 0.691 mg. Capsule cap composition: hypromellose – 22,080 mg, titanium dioxide dye (E 171) – 0.240 mg, iron oxide yellow dye (E 172) – 0.240 mg.

How to take, the dosage

Preferably take Linex® Forte capsules with a small amount of liquid during or after meals. Adults and adolescents over 12 years of age: 1 capsule 1 to 3 times a day. Children from 2 to 12 years old: 1 capsule 1 to 2 times a day. Infants and children under 2 years of age: 1 capsule 1 time a day. Diarrhea in children under 6 years of age should be treated under medical supervision. When prescribing Linex® Forte for children and patients who can not swallow the whole capsule, it should be opened, the contents poured into a spoon and mixed with small amount of liquid. The duration of treatment depends on the cause of dysbacteriosis and individual characteristics of the body. Linex® Forte can be taken until symptoms of dysbacteriosis disappear. Do not exceed the recommended daily doses of the drug.

Interaction

No undesirable interactions with other drugs were noted. When concomitant use of antibiotics, the recommended interval between the drug (antibiotic) and Linex® Forte capsules is 3 hours.

Special Instructions

Do not take Linex® Forte with hot drinks and do not take it with alcohol.

Patients should consult a physician if before starting the drug they have a body temperature above 38°C, blood or mucus in the stool, diarrhea lasts more than 2 days and is accompanied by severe abdominal pain, dehydration and weight loss; and if they have chronic diseases (diabetes mellitus, HIV infection).

The treatment of diarrhea requires reimbursement of lost fluids and electrolytes.

There is no need for special precautions when disposing of unused Linex® Forte.

Use in pediatrics

Diarrhea in children under 6 years of age must be treated under the supervision of a physician.

Influence on the ability to drive and operate machinery

The drug does not affect the ability to drive and operate machinery.

Contraindications

– hypersensitivity to the components of the drug;

– hereditary fructose intolerance, deficiency of sucrose, isomaltase or glucose-galactose malabsorption.

WARNING

With caution in diabetes mellitus (contains simple sugars), immune system deficiencies (e.g., HIV infection). Diarrhea in children younger than 6 years should be treated under the supervision of a doctor.

Side effects

Side effects of taking lactic acid bacteria are very rare (occur in 1 out of 10,000 people). Hypersensitivity reactions may occur.

Overdose

There are no data on the symptoms of overdose.
