Griseofulvin is an antifungal antibiotic.
It has fungistatic action.
It is active against dermatophytes of the genera Trichophyton, Microsporum, Epidermophyton.
Inhibits cell division of fungal cells in metaphase by disrupting the structure of the mitotic spindle.
Friseofulvin accumulates to varying degrees in skin, hair and nail cells that are precursors of keratin, making keratin resistant to fungal invasion.
As infected keratin separates, it is replaced by healthy tissue.

How to take, the dosage
Overly, during or after meals with vegetable oil.
In case of microsporiasis children are prescribed at the rate of 21-22 mg per 1 kg of body weight of the patient per day, adults – from 500 to 1000 mg (8 tablets per day).
The drug is taken daily until the first negative test for fungi, then in the same dose every other day for 2 weeks and then 2 times a week for 2 weeks.
In trichophytosis (superficial and infiltrative forms) and phavus in children – at the rate of 18 mg/kg of body weight according to the same scheme as in microsporiasis.
In case of onychomycosis, trichophytosis and scalp phavus the daily dose for adults with body weight less than 50 kg is 625 mg (5 tablets). Maximum daily dose is 1 g (8 tablets).
The daily dose in children and adolescents is 16 mg/kg body weight.
In case of onychomycosis the drug is prescribed daily during the first month, every other day during the second month and then 2 times a week in the same daily dose until the nails grow back.
The total duration of treatment is at least 8 months.
Interaction
Increases the effect of ethanol.
Decreases the activity of estrogen-containing oral contraceptives, indirect anticoagulants (due to induction of microsomal liver enzymes).
Barbiturates and primidone decrease antifungal activity.
Special Instructions
Treatment with Griseofulvin is performed under control of the morphological composition of the blood (once every 10-15 days).
Contraindications
Sensitivities, systemic lupus erythematosus, liver and kidney disease.
Side effects
CNS disorders: headache, dizziness, disorientation, unusual fatigue or weakness, insomnia, peripheral neuropathy, movement coordination disorders, paresthesias of extremities, confusion.
Gastrointestinal disorders: nausea, vomiting, diarrhea, gastralgia, candida stomatitis, hepatitis.
Hematopoietic disorders: granulocytopenia, agranulocytosis, leukopenia.
Allergic reactions: urticaria, skin rash, pruritus, angioedema, photosensitization, lupus-like syndrome, erythema multiforme exudative, Lyell’s syndrome.
