Fexofast has an antihistamine, anti-allergic effect. Fexofenadine hydrochloride is a non-sedative blocker of H1 – histamine receptors, it is pharmacologically active metabolite of terfenadine.

The antihistamine effect appears 1 hour after intake, reaches a maximum after 6 hours and lasts for 24 hours. When repeated use no development of tolerance has been noted. It does not have a choline- and adrenolytic and sedative effect. It does not cause changes of calcium and potassium channels function and QT interval in therapeutic doses.

Pharmacokinetics

After oral administration it is rapidly absorbed from the gastrointestinal tract, time after reaching maximum concentration (TC_max) is 1-3 hours. The mean value of maximum concentration (C_mah) after 180 mg administration is approximately 494 ng/mL, after 120mg administration – 427 ng/mL.

The binding to plasma proteins is 60-70% (predominantly to albumin and alpha1-glycoprotein). It does not penetrate through the blood-brain barrier. elimination half-life (T1/2) after multiple administration is 14.4 hours.

In patients with moderate (creatinine clearance 41-80 ml/min) and severe (11-40 ml/min) renal failure the T1/2 is increased by 59 and 72% respectively; in patients on hemodialysis the T1/2 is increased by 31%. Subjected (5% of dose) to partial extrahepatic metabolism. It is excreted mainly (80%) with bile, 11% – by kidneys unchanged.
