Pharmacodynamics
Cholecalciferol is a natural form of vitamin D formed in the skin by the action of sunlight. Compared with ergocalciferol 25% more active. Increases absorption of calcium in the intestine and reabsorption of phosphorus in the kidney tubules, normalizes the formation of the bone skeleton, helps maintain bone structure.
Calcium ions in the blood determine the maintenance of muscle tone of the skeletal muscles, myocardial function, promote nerve stimulation, regulate the process of blood clotting. It normalizes the functions of the perischymoid glands, the immune system, and affects the production of lymphokines.
Deficiency of vitamin D in food, impaired absorption, calcium deficiency, as well as lack of exposure to sunlight during rapid growth of the child leads to rickets; in adults – to osteomalacia; pregnant women may show symptoms of tetany, disorders of calcification of bones of newborns. An increased need for vitamin D occurs in women during menopause, as they often develop osteoporosis due to hormonal disorders.
Pharmacokinetics
Aqueous solution of vitamin D3 is absorbed better than the oil solution. Premature infants have insufficient bile formation and intestinal intake, which impairs absorption of vitamins in the form of oil solutions.
After oral administration it is absorbed in the distal small intestine. It is metabolized in the liver and kidneys. Passes through the placenta and into the breast milk. It is excreted in the urine and faeces. T1/2 is several days and increases in the case of renal insufficiency. It has the properties of cumulation.

Composition
One milliliter of solution (30 drops) contains 15,000 IU of cholecalciferol
(1 drop contains 500 IU of vitamin D3)
Excipients: Macrogoal glycerylicinoleate, sucrose 250 mg, diatrium phosphate dodecahydrate, citric acid monohydrate, banana flavoring, benzyl alcohol 15mg, purified water.

Special Instructions
Overdose should be avoided. Overdoses of vitamin D3 administered for long periods or shock doses can cause chronic hypervitaminosis D3. When determining the dose of vitamin D, the amount of vitamin D taken with food must be taken into account.
The determination of the child’s daily vitamin D requirement and the way it is taken is individualized by the doctor and monitored periodically, especially in the first months of life.
With caution should be used in patients with prolonged immobilization.
The simultaneous administration of high doses of calcium should be avoided.
The calcium levels in blood and urine should be monitored during treatment.
Contraindications
Overdose
Symptoms: decreased appetite, nausea, vomiting, constipation, restlessness, thirst, polyuria, diarrhea, renal colic. Headache and muscle and joint pain, mental disorders (depression), stupor, ataxia, progressive weight loss. Renal function disorder with albuminuria, erythrocyturia, polyuria, hyperkaluria, hypostenuria, nycturia, increased blood pressure. In severe cases – corneal clouding, edema of optic nerve papilla, iris inflammation (up to cataract development). Calcium nephrourolithiasis, systemic calcinosis may develop. Rarely, cholestatic jaundice.
Treatment: withdrawal of the drug. Symptomatic, supportive therapy. Taking plenty of fluids.
