Pharm Action:
Artrodarin is an anti-inflammatory drug, an inhibitor of the synthesis and activity of interleukin-1 (IL-1). IL-1 plays an important role in the development of inflammation, degradation and subsequent destruction of cartilage in osteoarthritis.
Diacerein also inhibits the action of other cytokines that cause inflammation, including IL-6, TNFα. Diacerein also inhibits the formation of metalloproteases (collagenase, elastase), which are involved in the process of cartilage damage. With long-term use diacerein stimulates the synthesis of proteoglycans and has no effect on the synthesis of prostaglandins.

Absorption and metabolism
After oral administration of Arthrodarin, diacerein is rapidly absorbed from the GI tract and is completely deacetylated to Rein. Administration of the drug simultaneously with food increases bioavailability by 25%. Cmax of reine in plasma is reached after 144 min. Cmax is 3.15 mg/l after a single dose of 50 mg. When multiple doses of the drug Cmax of Rein is increased due to cumulation.
Distribution and excretion
The binding of rein with albumin is almost 100%. Rein penetrates the HEB and the placental barrier. T1/2 is 255 min. Rein is excreted by the kidneys unchanged (20%) and as glucuronide (60%) and sulfate (20%).

How to take, the dosage
The standard dose is 1 capsule 2 times a day (morning and evening, after a meal).
The capsules should be swallowed whole, without chewing, and with water.
The effect of treatment should be expected after 2-4 weeks.
Artrodarin is taken continuously for a long time or in courses of at least 4 months.
Interaction
Artrodarin® should not be taken concomitantly with substances affecting the quality of intestinal contents and the rate of emptying (including large amounts of fiber).
Antacids decrease absorption of the drug.
When concomitant use with antibiotics or with chemotherapeutic drugs affecting the intestinal microflora, an increased incidence of adverse reactions from the intestine is possible.
Special Instructions
The duration of treatment can be long, depending on the result achieved. In the period preceding the development of the therapeutic effect.
Artrodarin can be taken simultaneously with analgesics and non-steroidal anti-inflammatory drugs.
Contraindications
– pregnancy;
– lactation (breast-feeding);
– childhood and adolescence (under 18 years);
– individual intolerance to the components of the drug Artrodarin or hypersensitivity to anthraxionic drugs (eg, laxatives).
Caution is prescribed to patients with irritation of the lower intestines, with chronic renal failure.
Side effects
In case of hypersensitivity to the components of Artrodarin there may be allergic reactions of varying degrees.

Overdose
