Allopurinol-Egis disrupts uric acid synthesis.
It is a structural analog of hypoxanthine. It inhibits the enzyme xanthine oxidase, which is involved in the conversion of hypoxanthine to xanthine and xanthine to uric acid.
This causes a decrease in the concentration of uric acid and its salts in the body fluids and urine, which promotes the dissolution of existing urate deposits and prevents their formation in the tissues and kidneys.
Allopurinol administration increases urinary excretion of hypoxanthine and xanthine.
Indications
Treatment and prevention of gout and hyperuricaemia of any genesis (including in combination with nephrolithiasis, renal failure, urate nephropathy). Increased formation of urate due to enzyme disorders.
Recurrent mixed oxalate-calcium kidney stones in the presence of hyperuricosuria.
Prevention of acute nephropathy in cytostatic and radiation therapy of tumors and leukemia, as well as in complete therapeutic fasting.

Composition
1 tablet contains 100 mg allopurinol;
auxiliary ingredients:
Lactose,
Starch,
Polividone,
talc,
magnesium stearate,
sodium amylopectin glycolate.
How to take, the dosage
Dosing regimen of Allopurinol-Egis is established individually, under control of urate and uric acid concentrations in blood and urine. In adults when administered orally – 100-900 mg/day, depending on the severity of the disease.
The frequency of intake is 2-4 times a day after a meal. Children under 15 years of age – 10-20 mg/kg/day or 100-400 mg/day.
Maximum dose: in kidney function disorders (including those caused by urate nephropathy) – 100 mg/day. Increase of this dose is possible in cases if during the carried out therapy increased concentrations of urate in blood and urine are saved.
Interaction
In concomitant use allopurinol increases the effect of coumarin anticoagulants, adenine arabinoside, and hypoglycemic drugs (especially with impaired renal function). Uricosuric agents and salicylates in high doses reduce the activity of allopurinol.
When allopurinol and cytostatics are used concomitantly, myelotoxic effects are more common than when used separately.
In concomitant use of allopurinol and azathioprine or mercaptopurine cumulation of the latter in the body is observed because due to inhibition of xanthine oxidase activity required for biotransformation of the drugs by allopurinol, their metabolism and elimination slows down.
Special Instructions
The daily amount of fluid intake is at least 2 liters.
Patients with tumor disease should be given a minimum dose before starting treatment with cytostatics.
In the beginning of gout treatment, an exacerbation of the disease may occur.
Please note that large urate stones may dissolve in the renal pelvis and then enter the ureter.
Contraindications
Developed liver and/or kidney function abnormalities, pregnancy, lactation.
High sensitivity to allopurinol.
Side effects
Nausea, diarrhea, skin rashes, and fever may occur.
Overdose
Symptoms: nausea, vomiting, diarrhea, dizziness, oliguria.
Treatment: forced diuresis, hemo- and peritoneal dialysis.
Pregnancy use
Allopurinol-Egis is contraindicated in pregnancy and lactation.
